Every other claim on this site traces back to this mechanism, so it is worth understanding properly — including where it stops being demonstrated and starts being extrapolated.
What nerve growth factor does
NGF is a protein that keeps neurons alive, encourages them to grow processes, and supports the repair of damaged nerve tissue. It is not speculative — it won a Nobel Prize. Low NGF is associated with neurodegeneration.
Hericenones and erinacines
Lion's mane produces two compound families that stimulate NGF synthesis. Hericenones are found mainly in the fruiting body — the white shaggy mushroom itself. Erinacines are found mainly in the mycelium, the root-like network. In laboratory cell studies, erinacines have raised NGF synthesis by up to 200%.
This split is the single most practical fact in the whole category, because it means fruiting-body and mycelium products are not interchangeable. They deliver different compounds. Most marketing pretends otherwise.
Where the evidence thins out
Almost all of the NGF work is in vitro — cells in a dish — or in animals. Two questions remain genuinely open in humans: whether enough of these compounds survive digestion, and whether they cross the blood-brain barrier in quantities that matter. Erinacines are small and lipophilic enough to make it plausible. Plausible is not proven.
This is the most over-stated mechanism in the supplement aisle. It is also real. The overstatement is in how confidently the leap from dish to human brain gets made.
Questions about this
Frequently asked
Does lion's mane really increase NGF in humans?
It increases NGF synthesis in laboratory cell studies, substantially — up to 200% in some in vitro work with erinacines. Whether that translates to meaningfully raised NGF in a human brain after oral consumption has not been definitively demonstrated. This is the honest state of the evidence.
What is the difference between hericenones and erinacines?
Hericenones come mainly from the fruiting body; erinacines from the mycelium. Both stimulate NGF, but they are different molecules with different properties — erinacines are smaller and more likely to cross the blood-brain barrier. A product made only from one part delivers only one of them.
Can lion's mane repair nerve damage?
Animal studies have shown accelerated peripheral nerve regeneration after injury. There is no adequate human evidence to support treating nerve damage with it, and nobody should substitute it for medical care.
